Plasma lipoprotein metabolism in lean and in fat
نویسنده
چکیده
Plasma lipoprotein metabolism was studied in vivo in two lines of chickens produced by selection for high and low plasma very low density lipoprotein (VLDL) concentration. Rates of VLDL secretion were measured by determining the rate of accumulation of triglyceride in the plasma after intravenous injection of anti-lipoprotein lipase antibody. The clearance of VLDL-triglyceride and its uptake into liver and adipose tissue was examined using radioactively labeled VLDL synthesized in vivo. The rate of VLDL secretion was about threefold higher in the high-VLDL line as compared to the leaner, low VLDL-line (6.7 vs 2.1 pmol VLDL triglyceride/h per ml of plasma). The clearance of VLDL from the circulation of the low VLDL line was much faster than that of the high VLDL line (tu2 of 3.7 and 13.6 min, respectively). The proportion of administered radiolabel taken up by the abdominal fat pad was substantially greater in the fat line than in the lean line (11.9 vs 4.876, respectively). Lipoprotein lipase acitivities in leg muscle and heart were consistently greater in the low-VLDL line and B-hydroxybutyrate concentrations in the plasma of the low-VLDL line were significantly greater than those in the high-VLDL line (0.86 vs 0.48 fimol/ml). The results show that the approximately tenfold difference in plasma VLDL concentration between lines is primarily due to markedly different rates of hepatic VLDL production and that selection has made a major effect on partitionin W e pmpose that a direction of fatty acids to oxida$ion rather than VLDL ayp&esis in the liver of birds of the low-VLDL line is amajm c;4uSe &their Iw rate of VLDL secretion and makes iin imppngaat cpritrikn&q.to improved o@ciency of protein utilization. متن کاملMetabolism of very low density lipoproteins in genetically lean or fat lines of chicken.
Metabolism of very low density lipoproteins (VLDL) has been compared in fat (FL) and lean (LL) lines of chicken. When refed after fasting, plasma triglyceride concentration reached a significantly higher plateau in FL, although their feed consumption was lower than in LL. Newly synthesized VLDL were studied using anti-lipoprotein lipase antibodies. VLDL triglyceride (TG) concentrations were inc...
متن کاملLipoprotein metabolism in the fat Zucker rat: reduced basal expression but normal regulation of hepatic low density lipoprotein receptors.
Hyperlipoproteinemia is one of the phenotypic characteristics of the fat Zucker rat that carries a mutation in the leptin receptor gene. In the present study, we studied the regulation of hepatic low density lipoprotein (LDL) receptor expression in lean and fat Zucker rats. Compared with lean rats, the fat ones had a pronounced (approximately 60%) reduction in hepatic LDL receptor expression, w...
متن کاملGenetic loci controlling body fat, lipoprotein metabolism, and insulin levels in a multifactorial mouse model.
We analyzed the inheritance of body fat, leptin levels, plasma lipoprotein levels, insulin levels, and related traits in an intercross between inbred mouse strains CAST/Ei and C57BL/6J. CAST/Ei mice are unusually lean, with only approximately 8% of body weight as fat, whereas C57BL/6J mice have approximately 18% body fat. Quantitative trait locus analysis using > 200 F2 mice revealed highly sig...
متن کاملChanges in Biochemical Parameters Related to Lipid Metabolism Following Lithium Treatment in Rat
Lithium is widely used in medicine as an anti-depressive drug. In spite of abundant literature, questions on the side effects of lithium ions are far from being answered. In this study, the effects of lithium on biochemical parameters related to lipid metabolism were investigated. Male Wistar rats were treated with different doses of lithium for a period of up to 60 days. Blood samples were col...
